As a quick rule of thumb, anything in the Desktop Cleanup folder can be safely deleted without compromising your system, because your computer and installed programs should be smart enough to not install critical things there.

Specifically, anything that says "shortcut to" or has an arrow in the lower left corner is a shortcut and is definately safe to delete. Deleting a shortcut doesn't touch the program it points to. Installation programs are safe to delete, they are not needed after the program is installed. Documents and stand-alone applications you'll have to determine on your own.
