As a mechanics wonk, the One-2-One system really caught my eye. I tend to work behind the screen so I was especially taken by the chapter on running the game. It is not long but has really solid advice for scenario creation and pacing to maintain the atmosphere. Overall the system is tight and interesting, well worth the cost of admission. The scenarios are top notch and brought a couple of designers onto my radar (Ruth Tillman, Chris Spivey). For more details, see my review on Amazon.
As an aside, in my experience it can be difficult to harmonize the schedules of 4-5 players for a 4+ hour gaming session. In that context, it seems there is a real opportunity to look into tuning not just scenarios but variants of existing systems to shine in one-on-one play.
