In the answer about a dwarf and an elf there is a piece of truth. I dare to say one of the mark of identities is the group of heroes with different classes and from diverse races/species. Usually in the standar fantasy there is one (human) main character and maybe his sidekicks, or the group is relativelly homogenous, fighters or spellcasters.

Now in some isekai mangas there are more diverse roles: tank fighter, stealth(rogue), magic buffer(cleric) and magic crystal canon(arcane spellcasters).

Other point is the worldbuilding. D&D is designed for "the monster of the week" while most of standar fantasy there is only one main antagonist faction. When this is defeated then the world becomes too peaceful for any possible sequel or spin-off.
