When determining the highest values of the defenses amongst the targets, are the target's bonuses and penalties to AC, Reflex, and Fortitude, taken into account? Or is it always the unmodified values of these defenses?

If I understand you correctly, you're asking if the fighter's Amulet of Protection +2 counts toward his Fort Defense for purposes of the power, or would it be his Fort Defense without the amulet?

My understanding is that you just take a look at what their defense scores are, with their gear, and use that. Though, to be honest, not sure how it works if a character has a temporary bonus (like from the Shield Power) up at the time Mantle is cast.

You take bonuses and penalties into account. There's really no such thing as an 'unmodified' defense, as a defense without any bonuses is always 10.
