Can a glave-wielding Cleric cast a spell when holding his glave?

Sure, just hold it in one hand to cast the spell, and resume your normal fighting grip afterwards.

I would certainly allow the cleric to hold it temp in one hand and cast a spell. However, I would not allow a player to have a large shield in one hand and a weapon in another to cast a spell without some metamagic feats.

Yes having him hold in in hand hand is fine, thus allowing him to cast the spell. Note, that if it's a full round casting or more, he would not be considered armed during this time.
