I don't know if I'd call it my best boss fight, but I'll tell you of my most recent one. The characters - Aldoth (dragonborn fighter), Armadillo (shifter monk), Dash d'Orien (human fighter), and Femur Dirtnap (high elf wizard) - have figured out the location of the suspected kidnapper of the scion of House Medani, a ruined temple of Aureon in the depths of Sharn. After fighting the villain's lieutenants - the gargoyle, Crag the Nose, a psychotic choker, Gasper Snatch, and a toothless bugbear thug called Smiley - in the vestibule and sanctum of the temple, they pressed on to encounter Mange, a shifter berserker, and Nipplespike, a hobgoblin monk festooned with body piercings, on the rickety, winding stairs leading up the bell tower. As they made their way up the crumbling stairs, Manger and Nipplespike threw javelins and hand axes at them. Armadillo and Dash closed the distance and took on Nipplespike while Femur hurled spells from the cover provided by a pile of rubble. Aldoth meanwhile fashioned a lasso and managed to snag Mange's outstretched arm and give it a yank, pulling her off the stairs to the hard ground 40 feet below. The lieutenants dispatched or routed, the PCs made it up the stairs to the belfry to face the master inquisitive of Sharn turned scheming villain, Dick Barrage. Zelina d'Medani was held fast to the bell with a strong steel chain and complex padlock. A powerful acid applied by Dick Barrage sizzled on the bolts holding the bell in place - if the PCs didn't rescue Zelina in three rounds, the bolts would give and the bell would plummet, taking her with it to the ground 80 feet below. After a suitably villainous monologue, the characters leaped into action. Dash and Armadillo rushed to engage Dick Barrage in a clash of swords and fists. Aldoth moved to secure a rope to Zelina. Femur used prestidigitation to cleanse the bolts of the acid eating away at them, buying the party some time. As Dick Barrage's sword cane rained blows upon Dash and Armadillo, it was clear the party was outmatched. Dash was on his last legs, desperately fighting against the master inquisitive atop the beam holding the bell in place as Dick tried to break the beam to drop the bell. Femur picked the padlock on the chain holding Zelina and Armadillo whisked her away to safety. His plans foiled, Dick Barrage lashed out at the PCs, disarming them and striking repeatedly. Short on options, the heroes focused on the beam, quickly smashing it. With a loud crack, it snapped, Dick Barrage slipped off, hitting the side of the bell - DONG! - before ultimately falling to his death. In the aftermath, the party licked their wounds and counted their blessings for it was truly a close call. The baron of House Medani rewarded them handsomely and the Sharn Inquisitive sang their praises on the front page.
