I very much commend the "Possible Lines" approach to defining NPC's in an adventure, mostly 'cos I've been doing it for years. ;)

Stick a few quotes from the character, both general and specific to one or more encounters, in your notes, and it's a great way to keep the characterisation sharp during your improv.
 



Since Gabe started posting about running his 4E game I've found it interesting reading: he approaches the task without decades worth of role playing preconceptions that many of us have built up by now.
