I have noticed since D20 became popular that alot companies and fans had a chance to created the most outragous of settings and interesting worlds. what I have is spac travel with your fantasy races, video-games in table-top, historical settings and even anime settings has been done.

The thing that I liked to see but have not are runes being the center of the campaign. Has anyone a seeting does this?

RuneQuest?

You want D20 Glorantha. I wish someone would make this!

Indeed, runes are a deep part of Greg Stafford's Glorantha (one of the three greatest secondary worlds created in any medium, alongside Middle-earth and Tékumel). You can go here and download the introduction to Gloranthan Runes from the amazing HeroQuest RPG.

As for d20 Glorantha, Greg is firmly agin it. (Though I don't think d20 is much less suited to Glorantha than RuneQuest is.)

If your looking for a rune based class, check out Arcana Unearthed. The Runethane is a great class to add runes to a campaign. The setting also involves runes, but not as a primary theme, more of a side effect of having a rune based class.
