A simple question...

What's in the fully fledged 224 page True20 Adventure Roleplaying rulebook that isn't in the more compact 160 page True20 Pocket Player's Guide?

According to this thread on the True20 forums, the principle difference are
the bestiary and world info is gone, along with the GMing hints, but assuming one had the core in PDF and had read those rules enough to remember ordinaries, minions, and the rules that affect them, it should be easy to run a True20 game with just the pocket guide and the True20 Bestiary.

So, you'll also miss out on the tips and tricks that make the system even more streamlined and useful, in addition to the example settings, feats and monsters. On the other hand it will have much of the errata and clarifications incorporated into it. This thread may also be of use.
