I'm a sucker for The Shadow Over Innsmouth as it's my favorite Lovecraft story of all time. I'm a little surprised there's no votes for The Call of Cthulhu yet. That's a good one too. Actually, all six of those are pretty good. My second favorite isn't an option here, Herbert West -- Reanimator.

Innsmouth seems like it would make the best game as there's a pretty gamable escape plot. Same with Dunwich Horror--find and disrupt Whateley's summoning.

As Deset Gled says, games and books are different media. Dreams in the Witch-House--are you going to kill Keziah Mason and Nyarlathotep? But I don't think anyone had mashed up non-Euclidean geometry and Salem witches before.
