How exactly was it an ass-pull?

We had strips that showed Roy getting the special spell disruption feat (although not the details of said feat) and a strip talking about the sword's special effects vs undead (again, without details).

That's foreshadowing (Chekov's Skill and Chekov's Gun), not an ass-pull.

The twist was that everyone presumed they'd be for battling Xykon, not vampire Durkon.

It is an ass pull for Durkula being knocked out of Hel's Might before it's duration is up, Roy's technique disrupts spellcasting, it doesn't end ongoing spells. it is an ass pull for deadly green energy, that's particularly harmful to undead, to suddenly send a sword's bearer into a ragestate. And it is an even larger asspull for that ragestate to grant a full heal.

I think that Roy's rage is what triggers the green energy. Being "particularly harmful to undead", it's likely to be positive energy, so it's no great leap for it to heal Roy in the process, or counteract spell effects that feed of negative energy.
