Help me Brainstorm: Fantasy Roanoke for Daggerheart

Doesn't have to be a time loop. It could even be something like a planeshift or planar convergence. The site where the colony was built is now in the legally-distinct Feywild. Maybe time has passed differently for them in this other plane. So about three generations have passed for them. They may even be part faerie or elf by now.
They built their colony in the summer, not realizing that the land is only there in the summer; in the other seasons it moves back into the Feywild.

And you don't want to be there when winter arrives.

Additional fun fact; the real big difference between Old World and New World historically, moreso than disease, is the presence of domesticable animals in the Old World (which pushed forward agriculturalism and thus cities and cities with lots of animals and thus lots and lots of disease)
That's not accurate. Domesticated animals is why Europeans had resistance to pox that first nations people didn't.
 

Ohhh, don’t forgot to show off turkeys. Everybody loves turkeys, they’re a NA thing, and there’s some thought that while the SW tribes had them fully domesticated already they may have been somewhat managed in the east cost region you’re set in.

Or make some subset of their weird ass clever Lurkeys like Dolmenwood, lol.
 

So I ended up deciding that the colonists had not showed proper deference to the forest goddess that owned the trees they used to build their homes, so she summoned them to her woods to serve her. The PCs learn this from a local shaman/druid and have gone to the forest to negotiate with her to get the colonists back. We broke just as they were entering the wood, so next time we will find out hat they can do to solve the problem.

That was exactly my point. Europeans had a lot of domesticated animals in close proximity to their living spaces which caused a lot of diseases which caused both (a) people with resistances to those diseases and (b) a lot of carriers of those diseases. The New World was relatively disease free, in large part because of the lack of domesticable animals.

All the New World had to give in return was syphilis, which Europeans spent centuries blaming each other for
