Spotting Invisibility?

How hard would it be for someone to Ready an action to attack (i.e. with arrows) an invisible target as soon as the target does something that might reveal its location (i.e. casts a spell like Melf's acid arrow that streaks out to a target)?

I'm assuming there would still be a 50% miss chance, but perhaps the square could be identified?

Yep, if you have a way to discern the square, i.e. since an attack originated from there, you should be able to pinpoint it exactly.

A spot check might still be required, since it might sound easier than it actually is, especially if you are looking in another direction (which the rules don't really cover).

By the rules the DC should be 20. Base DC of 0 and a +20 to pinpoint the location.

If the creature stays invisible (i.e. greater invisibility), then you still have the 50% miss chance, of course. That's because you can't see what direction the creature is ducking to and whatnot, even if you know where it stands.
