If you are referring to total defense (a Standard Action) and Open A Door (a Movement Action) then yes you can. The real question is has the thingy on the other side of the door made a listen check and is now aware of the person opening said door. You must determine suprise at this time to see whose actions go off first. The person opening the door may intend to open it using total defense but his action may occur after the beasties thereby catching him/her flat-footed.

It does not make sense outside of combat and won´t help you if you don´t win initiative... since you´d lose the dodge bonus anyhow and if you win initiative as soon as the door is open, you´ll be able to do something else.
