As long as the characters were acting as just that... characters, I'd say let the story play out. So what if one of the PCs bears a grudge against another of the PCs... in the right context that could be amazing. As long as that is what it is. What's not clear is the player motivation here...

I'd stay away from a spellcasting class. Lycanthropes lose the ability to cast while in animal/hybrid form, so that stinks. I don't have the books right in front of me, but the Dragon Shaman class has some healing abilities that aren't spells (it's in the PHB II... not sure if that is...

I couldn't find if it specifically forbidden, but I would rule that no, a caster's metamagic feats could not be applied to spell trigger / spell completion items. Gilladian's first point is specifically mentioned. That you may, at item creation, imbue a wand / scroll whatever with a spell...
