Jim if you want to generate random NPCs you could look at RPGAttitude's NPC Designer. The Game Ready Card gives you an index card with NPC information on it.

NPC Designer is a great product. Vascant is quick to respond to requests/corrections which is another big plus.

There's the Big List of RPG Plots for inspiration http://www.io.com/~sjohn/plots.htm. Most of those would do fine as a one-off game. You can set one of these up and add or remove encounters to speed things up or slow them down to make the whole thing fit the available time.
