I ran a couple highly successful Mage: The Ascention games for single players. In fact that's probably my favorite way to run most WoD stuff.
I'd say that the single most important thing becomes "What really interests the player?" Because they are going to be in the spotlight the entire time.
I've found that you can do things with a single player game that are very interesting, and which are very difficult in a game with 4 or 5 players. Things like lengthy investigations with very little combat, heavy roleplaying with NPC's that are directly involved with the single PC and even romantic relationships. It's much easier to portray how a love interest might act toward the PC when there aren't three other guys sitting around the room ready with a fart joke.
By the same token there are some things that are tough to do in solo games. You can't rely upon as wide a skillset when tackling challenges. And big combats are tough because it is too easy for a couple of bad die rolls to turn the whole thing against your protagonist.
I'd say that you should first figure out what genre she's most interested in, then figure out a general theme for the game and worry about system last.
