Nope, it is any attack that hits or misses. Thus you can use a wizard area attack you've multi-classed into...

Melee is only a requirement of the Interrupt Attacks you can do against enemies you've already marked.

At 11th (paragon lvl), if you have all the multiclass feats, you can "fully" multiclass to swap one of your at-will powers for an at-will power of the class you have multiclassed into.

Cleave only marks the target of the attack, not the cleaved target.

Suppose a Fighter uses his Cleave Attack, dealing damage to an enemy adjacent to the attack target. Can he mark that adjacent enemy as well?

No. Even if the adjacent enemy suffers damage, it was not the target of the attack, and cannot be marked.
