Ironically enough, I get that effect by not having 2 columns, because I never figured out how to do 2 columns in Word...

having the text be less dense will leave the space you need for notes, and probablly make it a little easier to find stuff. It will of course make more paper, which can mean more shuffling around. That's probably the trade-off of 2 columns vs. 1.
I suspect the 2 columns would be good for detailing all those locations, each room is short, and packing them together is less page flipping.
Non-location stuff, might be better suited to 1 column.
I could see doing an NPC in 2 columns (perhaps a 2 column, 1 row table) so you can put the lists of stuff in the right column (items, skills, feats), and the stats and fluff in the left column.
