Can two different character's "mark" the same target?

Short answer: no.

You can be subjected to only one mark at a time, and a new mark supersedes an old one.

Longer answer: No, but make sure that the effects you are considering are both marks. Things like Hunter's Quarry and Warlock's Curse are not marks. A Paladin's Divine Challenge includes a mark in it's description, so it is a mark (similar to a swordmage's aegis).

This is one problem with having 2 good defenders. They need to work together and plan ahead on who should be marking who. And, figure out ways to get their marks triggered.
