My "New World" campaign... I'm stuck!

Not with D&D magic, it won't. Remove disease is only a 3rd-level spell, and thus widely available, provided the OP uses relatively-normal demographics.

Start asking questions:
Who are the magic user? Elves, human, halflings, other?
Are they good or bad or? Blood magic, demon magic, nature, in hiding, from another plane?
Why are they there? A master race, running from something, protecting something

When you start answering the questions ideas should start coming to you, also check out the links in my sig.

Maybe it won't kill explorers, but unless you can cure everyone at once, and do it again and again, people will die.

What kind of mood are setting with this campaign? There's a great quote from William Burroughs about America thats a good mood setter : "Before the Mayflower, before the Indians, the Evil was here, waiting for us."

The Llama's on to something. By starting with the things you would see and feel if you were there you can then think of what kinds of life forms or creatures would have flourished there and then what kind of ecosystem would result. Also think about whther there are features that would be ideal for advance beings (outsiders or mage type). Would some flowerpot formations be ideal bases for a winged arcane using race?
 

Going with the "lost civilization" theme, check out Caverns of Thracia. It would fit right in. Also, your group may be the first settlers but surely rival nations are close on their heels. Conflict among the settlers themselves is almost inevitable. A rival faction could break off and be a source of conflict as they compete for the same resources.

Thanks for the pdf's, Frukathka! I haven't had time to give them a good read yet, but I gleaned them over once or twice and they look good. And you're quite bang-on with the Northern Crown ruleset... I may look into buying this farther down the line when my campaign is ready to evolve.
