It was a great series, and every book is worth a read. Still, though, the imagination went into the plot and characters, not the setting. Even then, each book had a completely new set of characters, often only keeping one from the previous book, and what setting there was was completely different in (each book takes place in a different epoch of man).

Each book, taken by itself, has some small measure of 'setting,' but certainly not enough to set an RPG in it. The series as a whole has no continuous or cohesive setting, however. That's just my observation, anyhow.

2001, 2061, 3001.

I can't remember if 2010 was a book before the movie.

Anyway, it has been done before, kind of, TSR did a 2001 (and 2010) supplement/module for Star Frontiers, oddly enough. I don't think it sold very well.

The trouble is, the world of 2001 is both outdated and yet too advanced compared to real life. It's got some hard science, but also stuff that so advanced it's magic.
 

2010 did come out first as a novel.

I realize they probably couldn't and wouldn't use anything from it, but I'll just pointlessly mention that there was also a monthly 2001 comic series from Marvel, written and penciled by Jack Kirby. It was like some insane cable anthology series, with the Monolith showing up every issue to change another person's life. It's mainly remembered for being where Machine Man first appeared; IIRC, he'd taken it over by the end of it's run.
