In what scenario is the number of fingers the primary deciding factor in adding a race to a campaign?

I'm building a spellcasting system where proficiency in a school of magic is dependent upon the total number of knucklebones present in the caster's hands. This of course has massive ramifications in the society's caste system, and political powerplays between different "handednesses" of the wizard academy's departments, not to mention the power leveraged by the Glove-makers Guild---

No, not really.

I'm not adding a race to a campaign. Just trying to find a bit broader justification for six-fingers on an NPC than random mutation or DM-fiat.

I liked your first explanation, I now wandering why you want a character with 6 rather than 5 fingers. Is it just aesthetic?
