Running a campaign in a large city

One of my favorite kinds of settings would be urban-based, city settings.

A lot of what I would say has already been echoed but a city based setting allows for a great deal of intrigue, plot and mystery. Further, one of the great opportunities a group has is interaction with villains, quasi-villains and others in a more civilized format.

This is also a great place to establish relationships with NPCs that allows for a lot of character development and role-playing. If that is a real passion for you and your players, this will be a great experience!
