I'll see your three questions, and raise you one more.
1. What one thing introduced in 4th Edition would you like to be included in the next edition of the game?
Rituals. Honourable mention to Skill Challenges (in concept if not implementation).
2. What one thing introduced in 4th Edition would you like to be excluded in the next edition of the game?
The continuous errata/updates (not strictly a 4e innovation - see 3e polymorph - but it's really gotten out of hand). Honourable mention to the micromanagement of short-term conditions and effects.
Oh, and dailies. At-will and encounter powers are fine, but as long as characters have a fixed number of dailies (and healing surges), the 15-minute adventuring day will always be a problem.

Skill ranks. Very much a taste thing, I know, but I prefer having more control over skills and than binary trained/untrained offered by 4e. (I'll happily admit that the 3e implementation leaves a lot to be desired.)
