Which DnD culture is the most likely to do this?

It seems to be a perfectly human thing.

Just like the party of paladin crusaders that will arrive and kill everyone in the colliseum, organisers, monsters, gladiators and spectators alike.

You could use illithids, but have the characters make their escape when the enclave is attacked by githzerai or githyanki (who probably don't care about the slaves one way or another). Also a good way to take the campaign planar if you want.
If your campaign is set in the Forgotten Realms, Skullport would be a good place to set it up. Slavery is legal, its underground, and pretty much has EVERY race on the planet involved in illegal activities.
If you REALLY wanna mess with the PC's, make them THINK they are deep underground. With the glare of torchlight they can never quite tell who (or what) is in the crowd (and they are probably too busy fighting off whatever is in the pit with them). Then, have them break out... right into the middle of a large metropolis.
 

I'd go with aboleths. Illithids would work well, but they seem too cold/alien to go in for pit fights. Aboleths would enjoy it more, and they'd be just as scheme-y. I get images of an arena and the small city that has grown up around it being situated in a gigantic, fungus-filled cavern where the walls and floor are honeycombed with aquatic passages - aboleths come there to enjoy the sights and sample the market that their tourist attraction is gathering for them. Besides, making the place appropriately damp would make it a more unique place to fight in. I could go for my party against a few skum hunting parties in an enormous colliseum, with gigiantic 'shrooms of every sort providing cover, atmosphere and the occasional fungoid monster that just sorta happened down there. And have the whole place knee-deep in water.

Oooh... I'm not sure about the rest... but having the arena full of small pools of water that are impossible to tell the depth of would add a lot to the fights. "Whoops, don't step too close, or you could get bull-rushed into the depths! Who knows what is waiting below!"

And of course, below, you need either lots of deadly game, or just one massive creature. Maybe both, but probably one or the other. Also helps keep the ring... "clean." I love this idea because I can make it work and it adds more depth to the whole sport... get it? Depth?
 

Alright, my current leanings are towards Duergar, but I've gotta ask the question:

"What race in Eberron would do this?" because I'm considering adding a lot of Eberron to the game, since we havn't really established the setting too thoroughly yet. We also have minotaurs from Dragonlance, and Draconians as options.
 

Alright, here is what I've decided on:

The coliseum is part of what was formerly the duergar empire. The Duergar have become slaves to the Elans (from the Psionics Handbook) and their cultists. The Elans are servants (also aberrations) of the mind flayers that once ruled the Duergar. The Duergar were formerly ruled by a group of especially crazy Derro. The Derro have been taken control of by the Elan telepaths, and now serve as tools for the manipulation of the kingdom.

Stone giants are also involved, they were formerly allied with the Duergar kingdom. The Elans will act as a sort of hook to relate all things to the mind flayers, while the Duergar will be the main obvious villain. So we have a stacked pile of manipulators pulling the cards.

The characters are all currently captive and being used for slave labor, but they are not guarded nearly well enough. Once they get past their mannacles and chains, it should be easy for them to overpower a guard or two and acquire some mundane weapons. Then I expect them to attempt to re-acquisition their gear. I'm going to try and keep 10-15 points of subduel damage on each character at a given time, so that they can be captured and thrown into the coliseum if they get into a mess they can't handle. If they do end up in the coliseum, I'm going to have a slave/gladiator stone-giant as the local champion, but he is only a gladiator because he KNOWS that the Derro are being manipulated by the Elans.

There should be plenty of opportunity for escape if they get outside of the city, but if they end up in the coliseum I've got a completely different plan for next session in which they will need to find their way out of the city (probably through an underground cavern, though I'm not "entirely" against them trying to really fight their way out if they are at 6th-7th level at that point, or even to bribe the guards). The group is currently 5th.

So what you's have to look for would be some race whith a a small nobility, a large amount of cannon fodder/common people. If they ever unite they could cause chaos, so the nobility keeps them happy with gladiatorial matches. You could probably go Orogs (also races of Faerun) who rule over their Orc minions. I like Orogs, they're smart Orcs.

I agree to this point of view but suggest goblinoids. The base high-numbered commoner is the goblin, while the bloodthirsty nobility is made up of hobgoblins. Hobgoblins are highly militaristic and organized. They could rule over a goblin populace working and suffering under them, then going to the gladiatorial matches for those who done well (as serfs and slaves). Hobgoblins themselves would greatly enjoy the games.
