The essential challenges are simple: You can't have an Assassin who isn't Evil.

And of course, the old problem of trying to recreate the flavor of a character outside the world that formed him.
Duplicate the assassin from the edition of your choice, rewrite without the alignment restriction (or change it to "any non-good"), tweak in the one or two other places required (as I recall, he's not a spellcaster, either), and done.

The rest are just specialized poisons, which I'd probably build using the potion creation rules.

It's not a hard challenge to recreate him.

You can't have an 'assassin' who isn't evil in the same vein you can't have a 'blackguard' that isn't evil. Because we have good versions of those archetypes that go by different names. Paladin, for example. In The Book of Ex Deeds pg. 73 there is the "Slayer of Domiel", effectively a "Good" assassin and is reasonably on par with the assassin. The SoD gets Divine Grace and Death Touch (instant death touch attack based on rolled d6s).

Absolutely you can.

Remember that the Assassin PrC is not the only way to be an assassin.

The Assassin PrC represents a member of a stereotypical fantasy Assassin's Guild - black magic, dark rituals, populace in fear, highest-bidder, etc.

An assassin is just someone who kills someone specific for a reason (which is often, but need not be, financial). Most PCs are, to a greater or lesser extent, assassins.
 


I don't know if there exist official poisons that cause bowel issues (Maybe a creative Bestow Curse?), but blindness, deafness, sickness, weakness, all have existing methods.

Ok, so I know I'm very biased, but this sounds like a Factotum to me.

Or a Rogue with good UMD. Perhaps a Wizard with some stealth abilities.
Ipecac is the closest I know. One round effect but since it was designed as a medicine, it has no save.

If you casted a Silent, Still Bestow Curse, creating your own curse such as Bowel irritation or impotence, would the target know the spell had been cast upon him/her?

Beguiler might make a great assassin like this as well.
