Creating New Base Classes

Sure. All the time (3.5). I do all kinds of customization and mixing and trading around class abilities, but I've also written new classes from scratch. Partially because archetypes I want weren't represented by existing rules, but it also helped me learn the game better even when I wasn't using them.

Yeah, I tried building classes in 4E, but it's a huge pile of work.

I've rebuild Star Wars Saga ed. from the ground up for a Final Fantasy game, though, with a lot of custom classes and rebuild/flavored base classes. And for 3.5 I redid a few of the psionic classes as well..
 

Never from scratch. Though in my 2E Darksun campaign I made so many adjustments to clerics and druids, they ended up being very different.
