In a fatansy campaign where such things as resurections and wishes are possible, there are in fact two main steps to an assasination rather than one:
1) Kill the target - this is the easier of the two and, of course, the most obvious of the steps
2) Remove the deceased target into isolation from possible help - this is where it gets tricky and the method of which depends entirely on the camapign situation
#2 can be done by removing the help from the target OR can be done by removing the target from the help.
Perhaps time the assassination for when the helpful cleric that is powerful enough to cast resurection or what not is gone on holy pilgrimage (though, have this fact stated in game well prior to the actual assination else it will seem to contrived if that fact is brought up after the death).
Or perhaps take the corpse away from the situation - hide the body in a locked room with wards against divination or toss the dead body into abag of holding, lug it around and toss him into the bottom of the forgotten seas...
Again, so many variations and possibilities for the second part of the assasination, it all depends on the campaign specifics (what's healing/helping resources are available to the target; what form the assinsation attempt will take; etc...)
