Thought about it a bit, and came up with a few things. I assume we're keeping it contemporary, 1980's - 2000's, so no Star Trek and such.
1) Indiana Jones is a definite, even though he may be a little old. You could probably explain his age away a little by the life-giving properties of the Holy Grail, as mentioned before. He represents the perfect likeable rogue, great charm, and doesn't have the "squeaky-cleanliness" that would get in the way of the Chaotic Good feel of what we're trying to accomplish here.
2) MacGyver. Obvious technical wizard, great improvisor. At the very least, he would be the main character in a list of secondaries.
3) Goliath (was thinking about him before I saw it posted

). Perfect for showcasing detriments in Humans, just like in the cartoon series (I still love that show). Fulfills the role of "monster character."
4) Optimus Prime. In a modern world, you're gonna have technical enemies, armies of robots, etc. Sooo..... combat that with one of your own. Also, we're talking 1980's Prime, not that new Armada stuff. Great chance for interaction with MacGyver on technical issues, and also with Goliath on issues related to "humanity."
5) Rat from The Core. Not a hero, not a villian, used by both sides. Offering equal prices, he'll side with the good guys. Perfect opportunity for an ambiguous hero/villian. Due to computer skills, and the state of the E-Conomy on the internet, it's a way to introduce the Nameless Corporate Villian.
6) A-Team's Face Man. 'Nuff said. Or, if you want more said, I'll say it. Disguises, infiltration, suave-ness, comedy. His playboy lifestyle could be a way to have bad things start, thus developing the beginning of a plot. Hitting on the BBEG's girlfriend wouldn't be looked upon too highly by said BBEG.
7) Kim(berly Anne) Possible. Pretty dang cool. Include her whole cast of sidekicks, especially Ron Stoppable and his pet naked mole rat Rufus. Appeals to the younger generation, great for "kewl" stuff, teenage/young adult issues, perfect comic relief.
8) One of the Predators. Mysterious, has his (its?) own agenda, opportunity to include Aliens as a villian.
Guest appearances by:
James Bond, the much more stylish British counterpart to Indiana. Head of the "European Branch" of the League? Also, gotta include Q (and watch for mixing Q with MacGyver). Marty McFly, mentioned earlier, would be wonderful. Then, you could include folks from other time periods; really gives free reign with story lines, as long as it isn't a cop-out way that is overused.
Jack Bauer (too "real-world" to be a main character) would be cool to see. He comes with several built-in plot hooks. How can anyone not say Fox Mulder?!? Bam, you've got government conspiricies, local shenanigans, alien (
Alien, even) invasions, and a host of wierd stuff to introduce. The guy from Early Edition, with the prophetic newspaper would be nice. "Look at this headline, Dr. Jones! 'Extroardinary League of Gentlemen Murdered!'"
