One way for a Paladin to mark multiple enemies is using the Divine Sanction property from the book Divine Power. Divine Sanction works like Divine Challenge except it only lasts till the end of your next turn and is used by attacking an enemy with a power that causes Divine Sanction (instead of requiring a minor action).

Divine Sanction lasts as long as the power granting Divine sanction says I believe. In other words, the duration varies.
