It depends on the group and the campaign, but all else being equal...
Controller: Tough one. After Arcane Power comes out*, maybe wizard (not sure on race). Although the idea of a deva invoker also sounds fun, but I haven't really seen if the mechanics will be as fun as the concept yet. Although, I'm really digging One Bad Egg's Witch Doctor class, but I'm feeding that interest with a multiclass right now.
Defender: Genasi swordmage. I really dig genasi. I really dig swordmages. They both fit perfectly together, so why not? Eventually, I might try my gnome fighter again. Originally ran him in 3.0 and the inspiration was the scene from Matrix with Neo going through the metal detector combined with the Throw Anything feat. Short swords and axes flew everywhere when he was around.
Leader: Bard. I'd probably go half-elf because I have a hard time NOT multiclassing, but that seems like such an obvious match it also turns me off of it at the same time. If I ever get a 4e Planescape campaign going, I have an old formian PC that would be perfect as a warlord (have to stat up the race, and that's formian, not fomorian!).
Striker: I've found this to be my favorite role, no contest. My ideal PC ever looks like it can finally be realized as a wild sorcerer multiclassed into dagger-focused rogue. The melee and close range carnage would be a complete joy. However, it's a bit indiscriminate, so in the campaign I'm running with my kids (and I have a DMPC to help out), I'm running a shadar-kai avenger (multiclass into witch doctor, because it's fun). Still magical strikery fun, but I don't have to worry about harming my kids' PCs by mistake which would get me into a lot of trouble.
* The first thing I did when I got the 4e PHB was stat out a wizard all the way to level 30, and then was sad when I realized that if I were to make a 2nd wizard, I probably wouldn't make any different choices. That lack of interesting (to me) options for what has always been a very customizable class really turned me off 4e for a while.
