Fluffwise, I like Dark Sun and the grittier tone. However, I think the Realms match up best with the mechanics of 4E and the ideals behind how the game is built.

For me, I like FR for it's Tolkien-esque look, Eberron's pulpy feel, and Dark Sun's Conan/Barsoom tone. They all have their own charm. I just wish that a Planescape, Spelljammer, and Ravenloft would've been more likely if not for the game's reliance on the default cosmology. Still, I'd love to see an innovative setting for 4e, something that's never been done before and could possible inspire other fantasy games, books, and movies for its look.
 

Of these, my favorite is probably PoL. It's vague enough that I can toss things in all willy-nilly, but still packed with thematic conflict.
It's funny: I like the mythology behind the PoL setting a lot more than I like the setting itself. The various settlements and lost nations of Nentir Vale don't really grab me - to a large extent, it doesn't feel "real" enough to me - but the mythology is great. Primordials? Gods? Fantastic!
I like the PoL cosmology and mythic history a lot - it's got great flavour and conflict built in (my contrast would be Greyhawk - a lot of flavour, but not so much conflict).

I don't use the Nentir Vale stuff. It seems harmless enough, but I started my 4e campaign with Night's Dark Terror and so far the campaign hasn't left that map.

I don't like FR. I've got nothing aginst the principle of Eberron but don't really want to run fantasy noir. I quite like the idea of Dark Sun, and if I run another 4e game will probably try it out.
