Hmm I don't think I can narrow it down. I'll list my top five, but they are all very close in my mind:

1) LOTR
2) Memory, Sorrow and Thorn by Tad Williams
3) The Dragonlance Chronicles by Weis and Hickman (very nice people in person too btw!)
4) The Icewind Dale trilogy and The Legacy by R. A. Salvatore
5) The Hobbit

I'm a bit disillusioned with series, after the direction Jordan's and Goodkind's have taken. Two or three books ago for each and both would have been on my list.

But the original Dragonlance chronicles and LOTR would of course be on my "best of" list. I'm also partial to the Shannara books, particularly the original trilogy.

Earthsea trilogy. excellent magic, a fine dungeon and great characters.

Fafhrd and the Mouser - superb setting with great characters.

REHoward Conan - the others may be fun but the best are the real thing.

We can only pick one? Awww, that's no fun...

Salvatore's Drizzt series (all of 'em except Spine of the World, the one that focused on Wulfgar) is good; I've read most of them twice, some of them three times. Looking forward to the continuation of the series.

The War of the Spider Queen is also great, and has a chance of becoming a favourite.

Weiss/Hickman's Dragonlance: Chronicles and Legends trilogies. Also enjoyed Death Gate Cycle and Star of the Guardians, both soloed by Weiss, IIRC.

Celia S. Freidman's: Coldfire Trilogy. Sweet!

Duh, how could I forget Harry Potter! It's cute, fun, and quick. Love the books.

However, to narrow it down to one series, I would have to choose the first Fantasy books that I ever read, the ones that got me addicted to the genre and the D&D game...
Raymond E. Feist's "Riftwar Saga."

I am more addicted to Harry Potter now than any series I've ever read. Before I would have said Elric, but reading bits of it now, I'm not sure why I was so into it. Maybe because I was a teenager and the angst appealed to me. Maybe it was the kewl sword.

Just to get the obvious out of the way....
Lord of the Rings
Now, on to the others.....

Drizzt Do'Urden, all of them!
The Deathgate series
The Serpentwar Saga
Harry Potter

And I know that this isn't fantasy or sci-fi, but I think they are awesome books and if you're at all into spies and military stuff, then definitely look into all of these....

Tom Clancy's Jack Ryan series (Patriot Games, Red October, Sum of All Fears....)

Stephen Donaldson's Gap series is way out in front for me.

If you discount sci-fi (or, more accurately, space opera), it gets harder.

The original Riftwar Saga (not the later stuff, and especially not anything he attached his name to but didn't actually write (Empress series, I'm looking at you  ), The Chronicles of Thomas Covenant and Song of Ice and Fire stand out from the crowd.

Guys, he was asking for JUST ONE SERIES OR NOVEL. That was the whole point of this question, to limit it to just ONE. He even went to the trouble of bolding the word one.

For me, it would be LOTR, hands down.
